Source text - English The two most commonly used chemically induced models of inflammatory bowel disease (IBD), murine dextran sulfate sodium (DSS) and 2, 4, 6-trinitrobenzene sulfonic acid (TNBS) colitis, are widely used for screening and assessing potential therapeutics due to the intestinal inflammation evoked through barrier destruction and immune imbalance. PF 06651600, aJanus kinase 3(JAK3) inhibitor, was prophylactically administered per os (PO) at various doses twice a day (BID) in order to block pro-inflammatory cytokines that contribute to IBD while preserving the signaling of regulatory cytokines shown to be protective in IBD. Intestinal disease was induced in the DSS model through the administration of 3% DSS in the drinking water tofemale C57Bl/6 mice on Days 0-7 followed by fresh water on Days 7-10. Intestinal disease was induced in the TNBS model, which ran until Day 2, 3, or 5, through a Day 0 intrarectal challenge of female Balb/c mice with 2 mg TNBS in a 40% ethanol solution.A PF 06651600 dose of 10 mg/kg BID was shown, in the majority of the chemically induced IBD studies, to improve disease through reductions in weight loss, disease activity, colon histopathology readouts of inflammation severity, and colon gene expression of biomarkers. Immunohistochemical (IHC) analysis highlighted the mechanistic action of PF-06651600, which showed decreased signal transducer and activator of transcription3 (STAT3) phosphorylation in the colon of mice treated with 10 and 30 mg/kg doses of PF 06651600, and STAT5 phosphorylation in the colons of mice treated with the 30 mg/kg dose of PF 06651600. Target modulation assays conducted on whole blood from these studies further supported the mechanistic action by which PF-06651600 showed efficacy; all doses of PF 06651600 decreased phosphorylation of STATs when stimulated by the gamma common chain cytokines IL-15 and IL-21 while having no effect on the phosphorylation of STATs when stimulated with IL-10or IL-27 which have been shown to have a protective effect in IBD.
