Objective: In this study we investigated the possible use of human demineralised dentine matrix (DHDM), obtained from the extracted teeth, as bone graft material and evaluated the expression of vascular endothelial growth factor (VEGF) induced by this material in the healing process of tooth sockets of rats.
Design: To evaluate bone regeneration and expression of VEGF induced by DHDM, thirty-two male Wistar rats weighing approximately 200 g were used. After maxillary second molar
extraction, the left sockets were filled with DHDM and the right sockets were naturally filled by blood clot (control). The animals were sacrificed at 3, 7, 14 and 21 days after surgery and upper maxillaries were processed for histological, morphometric and immunohistochemi-cal analyses. DHDM was used to evaluate the mechanical effect of bone graft material into sockets. Expression of VEGF was determined by immunohistochemistry in all groups.
Results: Our results demonstrated a significant increase in the newly formed bone tissue in sockets of 7, 14 and 21 days and a significant increase in VEGF expression at days 7 and 14 on treated sockets.
Conclusions: Our results showed that DHDM increases the expression of VEGF and accelerates the healing process in rats tooth sockets, by stimulating bone deposition and also vessels formation. These results suggest that DHDM has osteoinductive/osteoconductive potential and may represent an efficient grafting material on guided bone regeneration.
1. Introduction
Despite the high potential for regeneration of bone tissue, some repairs may be accompanied by intrabony defects so that and the repair of complex fractures and large bone defects, caused by several aetiological factors such as trauma, diseases, development deformities or tumour resection, often becomes frustrating.
One of the goals of skeletal tissue therapies is to accelerate the healing process, decreasing the period of bone maturation. Many strategies have been developed and tested to improve and accelerate bone repair, including the use of bone grafts and growth factors.2,3 However, classic bone grafting techniques, when used, rarely provide satisfactory results,4–6 and the use of growth factors may be infeasible because of the expensive costs.

Resumo
Objetivo: Neste estudo nós investigamos a possibilidade da utilização da matriz de dentina humana desmineralizada (DHDM), obtida a partir de dentes extraídos, usados como enxerto ósseo e avaliamos a expressão do fator de crescimento endotelial vascular (VEGF), que foi induzida por este material no processo de cicatrização de alvéolos dentários de ratos.
Método: Para avaliar a regeneração óssea e a expressão do VEGF induzida pelo DHDM, foram utilizados 32 ratos machos da raça Wistar, pesando aproximadamente 200 g. Após a extração do segundo molar superior, os alvéolos da esquerda foram preenchidos com DHDM e as alvéolos da direita foram naturalmente preenchidos com coágulo de sangue (de controle). Os animais foram sacrificados aos 3, 7, 14 e 21 dias após a cirurgia e os maxilares superiores foram processados para análise histológica, morfométrica e imunohistoquímica. O DHDM foi utilizado para avaliar o efeito mecânico do enxerto ósseo nos alvéolos. A expressão de VEGF foi determinada por imunohistoquímica em todos os grupos.
Resultados: Os resultados demonstraram um aumento significativo no tecido ósseo recém-formado nos alvéolos após 7, 14 e 21 dias e um aumento significativo na expressão de VEGF após 7 e 14 dias nos alvéolos tratados.
Conclusões: Nossos resultados mostraram que o DHDM aumenta a expressão de VEGF e acelera o processo de cicatrização em alvéolos dentários de ratos, ao estimular a deposição óssea e também a formação de vasos. Estes resultados sugerem que o DHDM possui um potencial osteoindutor/osteocondutor e pode ser eficiente na regeneração óssea assistida.
1. Introdução
Apesar do elevado potencial regenerativo do tecido ósseo, os processos de reparação podem causar alguns defeitos intraósseos. Desta forma, a reparação de fraturas complexas e defeitos ósseos maiores, causados por diversos fatores etiológicos como trauma, doenças, deformidades de desenvolvimento ou ressecção do tumor, muitas vezes é frustrada.
Um dos objetivos dos tratamentos de tecidos esqueléticos é acelerar o processo de cura, reduzindo o período de maturação óssea. Muitas estratégias têm sido desenvolvidas e testadas para melhorar e acelerar a reparação óssea, incluindo o uso de enxertos ósseos e fatores de crescimento. No entanto, as técnicas clássicas de enxerto ósseo, quando utilizadas, raramente fornecem resultados satisfatórios, e o uso de fatores de crescimento pode ser inviável devido aos custos elevados.
