Source text - English FIELD OF THE INVENTION
The subject invention relates to novel antimicrobial compounds, their compositions and their uses.
BACKGROUND
The chemical and medical literature describes compounds that are said to be antimicrobial, i.e., capable of destroying or suppressing the growth or reproduction of microorganisms, such as bacteria. For example, such antibacterials and other antimicrobials are described in XXXXXXXXXXXXXXXXXXXXXXXXX, editor, 1982), and XXXXXXXXXXXXXXX et al., XXXXXXXXXXXXXXXXXX 2d edition (1981).
The mechanism of action of these antibacterials vary. However, they are generally believed to function in one or more of the following ways: by inhibiting cell wall synthesis or repair; by alter¬ing cell wall permeability; by inhibiting protein synthesis; or by inhibiting synthesis of nucleic acids. For example, beta¬-lactam antibacterials act through inhibiting the essential penicillin binding proteins (PBPs) in bacteria, which are responsible for cell wall synthesis. As another example, quinolones act, at least in part, by inhibiting synthesis of DNA, thus preventing the cell from replicating.
The pharmacological characteristics of antimicrobials, and their suitability for any given clinical use, vary. For example, the classes of antimicrobials (and members within a class) may vary in 1) their relative efficacy against different types of microorganisms, 2) their susceptibility to develop¬ment of microbial resistance and 3) their pharmacological characteristics, such as their bioavailability, and biodistribution. Accordingly, selection of an appropriate antibacterial (or other antimicrobial) in a given clinical situation requires analysis of many factors, including the type of organism involved, the desired method of administration, the location of the infection to be treated and other considerations.
However, many such attempts to produce improved antimicrobials yield equivocal results. Indeed, few antimicrobials are produced that are truly clinically-acceptable in terms of their spectrum of antimicrobial activity, avoidance of microbial resistance, and pharmacology. Thus there is a continuing need for broad spectrum antimicrobials, which are effective against resistant microbes.

FUNDAMENTOS
A literatura sobre produtos químicos e medicina descreve compostos que se pensa serem microbicidas, isto é, capazes de destruir ou suprimir o crescimento ou a reprodução de microorganismos, como as bactérias. Por exemplo, esses bactericidas e outros microbicidas são descritos em Antibiotics, Chemotherapeutics, and Antibacterial Agents for Disease Control (Antibióticos, quimioterapeuticos, e agentes bactericidas para o controle de doença) (XXXXXXXXXXX, editor, 1982), e XXXXXXXXXXX et al., XXXXXXXXXXXXXXXXX (A base molecular da acção antibiótica) 2ª edição (1981).
O mecanismo de acção destes bactericidas variam. No entanto, acredita-se que no geral, funcionam em uma, ou mais, das seguintes formas: através da inibição da síntese da parede celular ou do reparo; através da alteração da permeabilidade da parede celular; através da inibição da síntese protéica; ou através da inibição da síntese de ácidos nucléicos. Por exemplo, os agentes bactericidas beta lactamas actuam através da inibição das proteínas de ligação da penicilina essenciais (PBPs) nas bactérias, as quais são responsáveis pela síntese da parede celular. Como outro exemplo, os quinolonas actuam, pelo menos em parte, inibindo a síntese do ADN, e dessa forma evitando que a célula se replique.
As características farmacológicas dos microbicidas e as suas compatibilidades variam para cada uso clínico específico. Por exemplo, as classes de microbicidas (e membros de uma classe) podem variar em 1) a sua eficácia contra vários tipos diferentes de microorganismos, 2) a sua susceptibilidade ao desenvolvimento de resistência microbiana e 3) suas características farmacológicas, tal como a sua biodisponibilidade, e biodistribuição. Consequentemente, a selecção de um bactericida adequado (ou outro microbicida) numa dada situação de teste clínico exige a análise de muitos factores, incluindo o tipo de organismo envolvido, o método de administração desejado, o local da infecção a ser tratado e outras considerações.
Entretanto, muitas outras tentativas para a produção de microbicidas aprimoradas, produzem resultados ambíguos. De facto, são produzidos poucos microbicidas que sejam verdadeiramente aceites clinicamente em termos do seu espectro de actividade microbicida, rejeição de resistência microbiana, e farmacologia. Dessa forma, existe uma necessidade contínua para microbicidas de amplo espectro, os quais são eficazes contra os micróbios resistentes.
