Alosteria significa "forma diferente". Enzimas alostéricas mudam de forma entre uma forma ativa e outra inativa como resultado da ligação dos substratos no sítio ativo ou com moléculas regulatórias em outros sítios.
No caso mais simples de enzima alostérica com uma forma ativa e uma inativa, a variação da velocidade da reação com o aumento da concentração do substrato é tipicamente uma curva em forma de S (sigmóide), diferente da curva cinética de Michaelis-Menten (hiperbólica).

Il che significa che ci sono almeno due siti in due differenti posizioni che sono capaci di legare dei leganti (substrati, inibitori...). La formazione di un legame di un legante ad un sito altera le proprietà dell'altro sito.
